UNIST, 미토콘드리아 전사 과정 규명 실마리 찾았다

김하진 교수팀, RNA 중합효소 '백트래킹' 현상 발견…"암·질환 치료 기대"

(울산=연합뉴스) 허광무 기자 = 울산과학기술원(UNIST) 연구진이 미토콘드리아(세포에 에너지 공급 역할을 하는 세포 내 소기관)의 DNA(유전정보) 전사 과정을 이해할 수 있는 실마리를 찾았다. 전사란 DNA 사본인 RNA(리보핵산)를 합성하는 과정을 말한다.

김하진 바이오메디컬공학과 교수팀은 미토콘드리아의 DNA를 전사하는 효소(RNA 중합효소)가 거꾸로 움직이는 현상(백트래킹)을 발견했다고 9일 밝혔다.

세포핵 전사에서는 이런 움직임이 발견됐지만, 미토콘드리아 전사에서 중합효소의 백트래킹이 발견된 것은 처음이다.

DNA에 저장된 유전정보는 전사 과정을 거쳐 단백질로 발현된다. 전사는 주로 세포핵에서 일어나지만, 미토콘드리아도 자체 전사 시스템으로 자신의 유전정보가 담긴 RNA를 합성한다. 이를 통해 단백질을 만들고, 합성된 단백질을 이용해 '세포 에너지 공급책' 역할을 수행한다.

미토콘드리아 전사 과정은 세포 생명 유지나 노화와 직결된다. 전사 과정을 정확히 밝힌다면 전사 과정을 교란해 암세포를 죽이거나, 전사 과정에 이상이 생겨 발생하는 질병을 치료할 수 있다.

RNA 중합효소는 유전정보 원본인 DNA 가닥 위를 한 걸음씩 전진하면서 복사본인 RNA를 합성하는 효소인데, 연구진은 이 중합효소가 DNA 가닥에 붙어 생기는 복합체의 움직임을 분석해 백트래킹을 암시하는 현상을 발견했다.

김 교수는 "이번 연구로 미토콘드리아 전사에서도 유사한 조절 메커니즘이 존재할 가능성이 커졌다"라면서 "미토콘드리아 유전자 발현을 조절하는 전사 경로를 이해하고, 미토콘드리아 관련 유전 질환 치료제와 항암제 등 개발에도 도움이 될 것"이라고 기대했다.

이번 연구는 국제 학술지 '네이처 커뮤니케이션즈'(Nature Communications) 8월 27일 자에 공개됐다.
